Product Specific Information

Mature rat IL-18 was expressed in Pichia pastoris, X-33. GenBank® Accession # P97636, amino acids 37 - 194. For protein verification, the expressed protein was N-terminal sequenced and begins with the following 10 amino acids: HFGRLHCTTA. Purity is >95% by SDS-PAGE analyses.

Activity: The biological activity of recombinant rat IL-18 was determined by its ability to induce IFNg production from KG-1 cells in the presence of recombinant mouse IL-12. 480 pg/mL and 610 pg/mL of IFNg were induced by 20 ng/mL and 40 ng/mL of recombinant rat IL-18, respectively.

Use: For in vitro applications. A dose-response assay should be performed to determine the optimal concentration for specific experiments.

Reconstitution and Dilution: Store lyophilized material at 4°C. Reconstitute in 0.5 mL of distilled or deionized water. Dilute in a buffer-containing carrier protein, aliquot, and store at or below -70°C, avoiding multiple freeze-thaw cycles. PBS with 1% BSA or a neutral media containing <10% FCS are suitable buffers.

Target Information

The protein encoded by this gene is a proinflammatory cytokine. This cytokine can induce the IFN-gamma production of T cells. The combination of this cytokine and IL12 has been shown to inhibit IL4 dependent IgE and IgG1 production, and enhance IgG2a production of B cells. IL-18 binding protein can specifically interact with this cytokine, and thus negatively regulate its biological activity.

For Research Use Only. Not for use in diagnostic procedures. Not for resale without express authorization.
