Thermofisher Scientific works with a variety of human tissue sources, including Tissue and Organ Procurement Organizations, qualified Research Tissue Organizations and prominent academic and medical centers through collaborations that follow rigorous regulations, certifications and/or accreditations (Source Facilities). Tissues obtained through source facilities are consistent with the applicable legal and ethical practices of the United States.
Source Facilities are required by federal law to maintain accurate records and donor confidentiality as well as adhere to certain applicable laws governing tissue used for research globally. These include, but are not limited to:
- 45 CFR Part 46, subparts A, B, C, D where applicable (US federal guidance on the use of human subjects in research)

The human liver cells are derived from tissue obtained from accredited institutions. Consent is obtained by these institutions from the donor or the donor’s legal next of kin, for the use of the tissue and its derivatives for research purpose.
We have used a ratio between 10:1 and 3:1 for 3D-spheroid hepatocytes to stellate cells for a successful 3D co-culture. We would recommend testing different ratios as the cells may vary from one type of model to another.
Yes, we have done co-culturing of hepatic stellate cells along with 3D-spheroid hepatocytes.
We are in the process of testing the metabolism of multiple low-turnover compounds using hepatic spheroids. We have found that a single spheroid containing 3,000 cells can be used for metabolic studies of high-turnover compounds.
We have found that hepatic 3D spheroids can be functionally viable for up to 28 days.
We recommend changing 50% of the media every 48-72 hr. You can either discard the media very carefully, using a multi-channel pipette, without disturbing the spheroids, or a programmable plate washer can be used.
Cryopreserved 3D-Spheroid Qualified Human Hepatocytes are primary hepatocytes isolated from a single donor that can form 3D-spheroids using Nunclon Sphera Plates within 5 days of seeding. We are not sure why every batch of human hepatocytes does not form 3D-spheroids. We speculate that this is a result of donor-to-donor variability and variation in the quality of tissue.
