SP1 Antibody (MA5-49142) in ICC/IF

Immunocytochemistry analysis of SP1 in Hela cells. Enzyme antigen retrieval was performed using IHC enzyme antigen retrieval reagent for 15 mins. The cells were blocked with 10% goat serum. Samples were then incubated in SP1 Monoclonal antibody (Product # MA5-49142) using a dilution of 5 μg/mL. DyLight®550 Conjugated Goat Anti-Mouse IgG was used as secondary antibody at 1:100 dilution and incubated for 30 minutes at 37°C. The section was counterstained with DAPI. Adding 0.2 mL of distilled water will yield a concentration of 500 µg/mL.

Positive Control - WB: human Hela whole cell, human Caco-2 whole cell, human Jurkat whole cell. IHC: human colorectal adenocarcinoma tissue, human glioma tissue, human laryngeal squamous cell carcinoma tissue, human lung cancer tissue, human ovarian cancer tissue, human thyroid cancer tissue, human tonsil tissue. ICC/IF: Hela cell. Flow: A431 cell.|Store at -20°C for one year from date of receipt. After reconstitution, at 4°C for one month. It can also be aliquotted and stored frozen at -20°C for six months. Avoid repeated freeze-thaw cycles.

Target Information

SP1 is a transcription factor that can activate or repress transcription in response to physiological and pathological stimuli. Binds with high affinity to GC-rich motifs and regulates the expression of a large number of genes involved in a variety of processes such as cell growth, apoptosis, differentiation and immune responses. Highly regulated by post-translational modifications (phosphorylations, sumoylation, proteolytic cleavage, glycosylation and acetylation). Binds also the PDGFR-alpha G-box promoter. May have a role in modulating the cellular response to DNA damage. Implicated in chromatin remodeling. Plays a role in the recruitment of SMARCA4/BRG1 on the c-FOS promoter. Plays an essential role in the regulation of FE65 gene expression.
