MBD1 Antibody (MA1-41025) in ICC/IF

Immunocytochemistry analysis of MBD1 in NIH3T3 cells. Samples were incubated with MBD1 monoclonal antibody (Product # MA1-41025) using a dilution of 2 µg/mL overnight at 4°C followed by anti-mouse DyLight 488 (Green) at a dilution of 1:500. Cells were fixed for 10 minutes using 4% PFA and then permeabilized for 5 minutes using 1X PBS + 0.5% Triton-X100. Actin was detected with Phalloidin 568 (Red) at a 1:500 dilution. Target Information

Methyl-CpG-binding domain protein 1 (MBD1) is a member of a family of mammalian proteins that specifically bind methylated regions of DNA. It is a transcriptional repressor that binds CpG islands in promoters where the DNA is methylated at position 5 of cytosine within CpG dinucleotides. Binding is abolished by the presence of 7-mG that is produced by DNA damage by methylmethanesulfonate (MMS). MBD1 acts as a transcriptional repressor and plays a role in gene silencing by recruiting AFT7IP, which in turn recruits factors such as the histone methyltransferase SETDB1. Isoform 1 and isoform 2 can also repress transcription from unmethylated promoters.
