CNR2 Antibody (PA1-744) in WB

Western blot analysis of Cannabinoid Receptor II was performed by loading 25 µg of PC-3 (lane 1) and Jurkat (lane 2) cell lysates onto an SDS polyacrylamide gel. Proteins were transferred to a PVDF membrane and blocked at 4°C overnight. The membrane was probed with a Cannabinoid Receptor II polyclonal antibody (Product # PA1-744) at a dilution of 1:1000 overnight at 4°C, washed in TBST, and probed with an HRP-conjugated secondary antibody for 1 hr at room temperature in the dark. Chemiluminescent detection was performed using Pierce ECL Pl... View More {{ $ctrl.currentElement.advancedVerification.fullName }} validation info. View more

Product Specific Information

PA1-744 detects cannabinoid receptor 2 (CB2) from human tissues.

PA1-744 has been successfully used in Western blot, FACS, immunocytochemistry and immunofluorescence procedures. By Western blot, this antibody detects an ~60 kDa band from human spinal cord homogenate, and a ~40 kda band from RBL2H3 cells, representing the different glycosylated forms of CB2.

PA1-744 antigen is a fusion protein containing the first 33 amino acid residues from human CB2.

Target Information

Cannabinoids exert their well known physiological effects through two G protein coupled receptors, cannabinoid receptor 1 (CB1) and CB2. Both cannabinoid receptors have been shown to inhibit adenylyl cyclase as well as stimulate the mitogen-activated protein kinase, MAPK. CB1 receptors also modulate ion channels through direct G-protein interactions. Delta 9-tetrahydrocannibinol and related ligands likely exert their psychoactive effects by inhibiting presynaptic N- and P/Q type calcium channels. CB2 is thought to function primarily in the immune system although it has been suggested to be present in the central nervous system, including the retina.
