CD163 Antibody (H088T03B06-A) in Flow

Normal human peripheral blood cells were unstained (left) or stained with CD163 Monoclonal Antibody, NovaFluor Blue 610-70S (right). All cells were co-stained with CD11b Monoclonal Antibody, Brilliant Ultra Violet 805 (BUV805) (Product # 368-0118-42). Total viable cells in the lymphocyte gate were used for analysis, as determined by LIVE/DEAD Blue (Product # L34962). Description
This MAC 2-158 monoclonal antibody reacts with human CD163, a high affinity macrophage scavenger receptor for hemoglobin and haptoglobin.

This product contains 1 vial of NovaFluor conjugate and 1 vial of CellBlox Plus Blocking Buffer.

Applications Tested
This MAC 2-158 antibody has been pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. This may be used at 5 µL (0.4 µg) per test. A test is defined as the amount (µg) of antibody that will stain a cell sample in a final volume of 100 µL. Cell number should be determined empirically but can range from 10^5 to 10^8 cells/test.

Master mixes
• Master mixes of NFs should be made at 2-8 °C and may be made up to 4 hours ahead of time.
• We do not recommend storing master mixes containing NovaFluor conjugates overnight or longer.

Whole Blood compatibility
• When utilizing whole blood (as opposed to density-gradient-purified PBMC), we recommend lysing red blood cells in bulk prior to staining with NovaFluor conjugates.
• See the Bulk Lysis of Human Whole Blood protocol here.
• Staining of whole blood with NovaFluor conjugates followed by lysis of red blood cells may result in higher-than-expected background staining.

Viability dye compatibility
• NovaFluor dyes are not compatible with DNA intercalating viability dyes.
• Do not use viability dyes such as propidium iodide, 7-actinomycin D (7-AAD) and DAPI. Invitrogen LIVE/DEAD Fixable Dead Cell stains are recommended for use with NovaFluor dyes.

CellBlox Plus Blocking Buffer
• This NovaFluor conjugate comes with CellBlox Plus Blocking Buffer (Cat. No. C001T03F01), essential for optimal staining.
• Use CellBlox Plus Blocking Buffer in all experiments with NovaFluor conjugates.
• Add 5 µL per sample to antibody cocktails/master mixes (regardless of how many Novafluor-conjugated antibodies are present) before combining with cells.
• CellBlox Plus Blocking Buffer is compatible with either Super Bright Complete Blocking Buffer or Brilliant Stain Buffer and can be used in antibody cocktails/master mixes with those reagents.
• For single-color controls, use 5 µL of CellBlox Plus Blocking Buffer per 100 µL of cell sample (10^3 to 10^8 cells).

Target Information

CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein, a member of the scavenger receptor cysteine-rich superfamily, and a receptor for the hemoglobin-haptoglobin complex. CD163 protects tissues from free hemoglobin-mediated oxidative damage, and may play a role in the uptake and recycling of iron, via endocytosis of hemoglobin/haptoglobin and subsequent breakdown of heme. CD163 is expressed exclusively on the cell surface of human monocytes and macrophages that evolve predominantly in the late phase of inflammation. Specifically, CD163 is present on all circulating monocytes and most tissue macrophages except those found in the mantle zone and germinal centers of lymphoid follicles, interdigitating reticulum cells and Langerhan's cells. CD163 is present on all CD14 positive monocytes, most CD64 positive monocytes, and shows higher expression on CD16 positive monocytes. CD163 is upregulated on mononuclear phagocytes by IL-10, IL-6 and dexamethasone. Lipopolysaccharide (LPS) and phorbol myristate acetate (PMA) both induce shedding of CD163 from the cell surface into plasma or cell supernatant. CD163 binds hemoglobin/haptoglobin complexes in a calcium-dependent and pH-dependent manner, and exhibits a higher affinity for complexes of hemoglobin and multimeric haptoglobin of HP1F phenotype than for complexes of hemoglobin and dimeric haptoglobin of HP1S phenotype. Further, CD163 also induces a cascade of intracellular signals that involves tyrosine kinase-dependent calcium mobilization, inositol triphosphate production and secretion of IL6 and CSF1.
