Product Specific Information

Description: IFN-alpha2 is a type I interferon, previously known as B-cell interferon, leukocyte interferon, lymphoblast interferon, and pH2-stable interferon. IFN-alpha2 is one of at least 23 different known variants of IFN-alpha. The individual proteins have molecular masses between 19-26 kDa and consist of proteins with lengths of 156-166 and 172 amino acids. IFN-alpha forms are produced by monocytes/macrophages, lymphoblastoid cells, fibroblasts, and a number of different cell types following induction by viruses, nucleic acids, glucocorticoid hormones, and low-molecular weight substances (n-butyrate, 5-bromodeoxyuridine). IFN-alpha2 demonstrates antiviral, antiparasitic, antiproliferative activities.

Applications Reported: The recombinant mouse IFNa2 has been reported useful for bioassay.

Applications Tested: The recombinant mouse IFN alpha2 has been tested for inhibition of the cytopathic effect of EMC virus on L929 cells. The ED50 for this effect is typically 0.1 ng/mL, corresponding to a specific activity of 1 x 10e7 Units/mg.

Source: E. coli expressed amino acids Cys24-Glu190 of mature mouse IFN alpha2 accession # NM_010503.

Bioactivity: The recombinant mouse IFN alpha2 has been tested for inhibition of the cytopathic effect of EMC virus on L929 cells. The ED50 for this effect is typically 0.1 ng/mL, corresponding to a specific activity of 1 x 10e7 Units/mg.

Endotoxin: Less than 0.01 ng/ug cytokine as determined by the LAL assay. Purity: >98% as determined by SDS-PAGE.

Molecular Weight: The protein has a predicted molecular mass of 19,361. The non-reduced protein migrates as a 16 kDa polypeptide on SDS-PAGE. The DTT reduced protein migrates as an 18 kDa polypeptide.

Storage and handling: For best recovery, quick-spin vial prior to opening. Use in a sterile environment.

Purity: Greater than 90%, as determined by SDS-PAGE.

Aggregation: Less than 10%, as determined by HPLC.

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

IFN alpha (Interferon alpha) is a Type I interferon belonging to a family of structurally related, species-specific proteins exclusive to vertebrates. The interaction of IFN-alpha with its receptor component result in the activation of a number of signaling pathways which are mainly involved in innate immune response against viral infection. In humans, the gene for IFN alpha is located on on chromosome 9. The type I interferons consist of 14 different alpha isoforms (subtypes with slightly different specificities), and single beta, omega, epsilon and kappa isoforms. Homologous molecules are found in many species, including rats and mice (and most mammals), and have been identified in birds, reptiles and fish species. IFN alpha is produced primarily by plasmacytoid dendritic cells, is a potent component of the anti-viral innate immune response and modulates adaptive immunity. Diseases associated with the dysfunction of IFN alpha include Hepatitis C and Hepatitis.
