About This Kit

The Human Soluble CD80 (Hu sCD80) ELISA quantitates Hu sCD80 in human serum, plasma, cell culture supernatants, or other body fluids. The assay will exclusively recognize both natural and recombinant Hu sCD80.

Principle of the method

The Human sCD80 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the binding of the second (detector) antibody to the target on a different epitope from the capture antibody. An antibody conjugated with enzyme binds the formed sandwich. After incubation and washing steps to rid the microplate of unbound substances, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen.

Target information

CD80 (B7-1) and CD86 (B7-2) are ligands of T cell critical costimulatory molecule CD28, and of an inhibitory receptor CTLA-4 (CD152). Both B7 molecules are expressed on professional antigen-presenting cells and are essential for T cell activation, and both molecules can also substitute for each other in this process. CD80 is rapidly induced on the surface of in vitro activated B cells, with Epstein Barr Virus (EBV) transformed B cell lines, with Burkitts lymphoma cell lines, with freshly isolated follicular B lymphoma cells, T cells, and monocytes. The B-lymphocyte activation antigen B7-1 provides regulatory signals for T lymphocytes as a consequence of binding to the CD28 and CTLA4 ligands of T cells. Diseases associated with CD80 dysfunction include gallbladder squamous cell carcinoma and myocarditis.
