Recommended positive controls: human peripheral blood lymphocytes.

In Neutralization studies, at 0.5-1.0 µg/mL, this antibody will block 50% of the bioactivity due to 0.3 µg/mL recombinant human sTNF RII, in the presence of 0.25 ng/mL TNF-alpha in a crystal violet cytolytic assay using mouse L929 cells. Anti-Human sTNF RII is tested for its ability to neutralize the biological effect of sTNF-RII but not sTNF RI in a cytolytic assay using mouse L929 cells, in the presence of TNF-alpha. The ND50 of the antibody is defined as the concentration of antibody resulting in a one-half maximal inhibition of the soluble receptor activity on a responsive cell line, when the soluble receptor is present at a concentration just high enough to elicit a maximum response.

Target Information

TNFR2 (tumor necrosis factor receptor superfamily member 1B) is a receptor with high affinity for TNFSF2/TNF-alpha. It has a much lower affinity for homotrimeric TNFSF1/lymphotoxin-alpha. The TRAF1/TRAF2 complex recruits the apoptotic suppressors BIRC2 and BIRC3 to TNFRSF1B/TNFR2. TNFR2 mediates most of the metabolic effects of TNF-alpha.
