IL-10 Antibody (MM011) in WB

Western blot was performed using Anti-IL-10 Monoclonal Antibody (16E3) (Product # MM011) and a 18kDa band corresponding to Interleukin-10 was observed across in Mouse rec. IL10. Human rec. IL10 was used to examine species cross reactivity. 250 ng Human rec. IL10 (Lane 1), Mouse rec. IL10 (Lane 2) were electrophoresed using NuPAGE™ 12% Bis-Tris Protein Gel (Product # NP0341BOX). MM011 targets IL-10 in ELISA applications and shows reactivity with mouse samples.

The MM011 immunogen is recombinant mouse IL-10.

MM011 detects IL-10 which has a predicted molecular weight of approximately 19 kDa.

This antibody is produced by injecting Rat IgG secreting hybridoma cells into the peritoneum of mice. The resulting ascites is collected from the mice and the antibody is purified.

This product is a Low Endotoxin formulation.

This product has been tested for endotoxins by limulus amoebocyte lysate (LAL) assay and contains an endotoxin concentration of less than or equal to 10 endotoxin units per milligram (EU/mg).

Target Information

IL-10 encodes a protein that acts as a cytokine and is primarily produced by monocytes, with some production by lymphocytes. This cytokine has various effects on immunoregulation and inflammation, including down-regulating the expression of Th1 cytokines, MHC class II Ags, and costimulatory molecules on macrophages. It also enhances B cell survival, proliferation, and antibody production. Additionally, IL-10 can block NF-kappa B activity and is involved in the regulation of the JAK-STAT signaling pathway. Knockout studies in mice have suggested that this cytokine is an essential immunoregulator in the intestinal tract. Mutations in this gene have been associated with an increased susceptibility to HIV-1 infection and rheumatoid arthritis. Diseases associated with IL10 include Graft-Versus-Host Disease and Human Immunodeficiency Virus Type 1.
