Preparation: Produced from sera of rabbits immunized with highly pure Recombinant Human Adiponectin. Anti-Human Adiponectin-specific antibody was purified by affinity chromatography and then biotinylated.

Sandwich ELISA: To detect Human Adiponectin by sandwich ELISA (using 100 µL/well) a concentration of 0.25-1.0 µg/mL of this antibody is required. This biotinylated polyclonal antibody, in conjunction with PeproTech Polyclonal Anti-Human Adiponectin (500-P239) as a capture antibody, allows the detection of at least 2000-4000 pg/mL of Recombinant Human Adiponectin.

Western Blot: To detect Human Adiponectin by Western Blot analysis this antibody can be used at a concentration of 0.1-0.2 µg/mL. When used in conjunction with compatible secondary reagents the detection limit for Recombinant Human Adiponectin is 1.5-3.0 ng/lane, under either reducing or non-reducing conditions.

Target Information

Adiponectin is a protein hormone secreted exclusively by adipocytes that regulates the metabolism of lipids and glucose. Adiponectin has been shown to play a role in various physiological processes including homeostasis and obesity. Decreased levels of adiponectin are associated with obese individuals as well as insulin resistance and hyperinsulinemia (type 2 diabetes). In mice adiponectin has been shown to improve insulin signaling, thus ameliorating insulin resistance. Through activation of AMP kinase and PPAR-alpha, adiponectin can increase fatty acid transport, oxidation, and dissipation in skeletal muscle, leading to a reduction of intramyocellular lipid levels and thus improved insulin signaling.
